Branched-chain amino acids and the association with type 2 diabetes

چکیده
The prevalence of type 2 diabetes is soaring worldwide and is now recognized as one of the main threats to human health being associated with comorbidities, such as cardiovascular disease. Type 2 diabetes is a condition characterized by abnormalities in carbohydrate, lipid and protein metabolism, with the most characteristic features being hyperglycemia and dyslipidemia. The underlying pathological aberrations comprise insulin resistance and bihormonal dysfunction of the pancreatic aand b-cells. As aforementioned, high-protein diets are associated with impaired glucose tolerance, insulin resistance and an increased incidence of type 2 diabetes. Protein consists of amino acids (AAs). AAs were traditionally classified as essential or nonessential for humans and animals. Essential AAs cannot be synthesized from other compounds in the body at the level required for normal growth, so they must be obtained from food. Leucine, isoleucine and valine are named as branchedchain amino acids (BCAAs). BCAAs are the most abundant of the essential AAs. Leucine is the most abundant BCAA in many dietary proteins, accounting for over 20% of total dietary protein obtained from the human diet. Of the AAs studied, the BCAAs have generated the most research interest, as they have emerged as potential biomarkers of metabolic disease. Circulating levels of BCAAs are elevated in individuals with obesity, impaired fasting glucose and type 2 diabetes. Furthermore, circulating levels of BCAAs have the potential to predict development of type 2 diabetes.

Background: Diabetic nephropathy is a chronic kidney disease and of more common complications of type 2 diabetes mellitus. The current diagnostic markers of diabetic nephropathy, albumin and creatinine, are only able to catch the disease in the stage of renal damage. The aim of this study is evaluation of targeted metabolomics of serum amino acids to identify the association of the changes of s...

Background& purpose: Today, among athletes, the use of amino acid supplements with the aim of reducing muscle injuries is more important than in the past. the aim of this study was to survey the effect of branched chain amino acids (BCAA) consumption before exhaustive exercise along with 1-month vitamin E supplementation on lactate dehydrogenase (LDH) and creatine kinase (CK) levels of active f...
متن کاملBranched-Chain and Aromatic Amino Acids Are Predictors of Insulin Resistance in Young Adults
OBJECTIVE Branched-chain and aromatic amino acids are associated with the risk for future type 2 diabetes; however, the underlying mechanisms remain elusive. We tested whether amino acids predict insulin resistance index in healthy young adults. RESEARCH DESIGN AND METHODS Circulating isoleucine, leucine, valine, phenylalanine, tyrosine, and six additional amino acids were quantified in 1,680...
متن کاملGenetic Predisposition to an Impaired Metabolism of the Branched-Chain Amino Acids and Risk of Type 2 Diabetes: A Mendelian Randomisation Analysis
BACKGROUND Higher circulating levels of the branched-chain amino acids (BCAAs; i.e., isoleucine, leucine, and valine) are strongly associated with higher type 2 diabetes risk, but it is not known whether this association is causal. We undertook large-scale human genetic analyses to address this question. METHODS AND FINDINGS Genome-wide studies of BCAA levels in 16,596 individuals revealed fi...
